The World Health Organization has identified antimicrobial resistance (AMR) as an imminent global health crisis requiring urgent development of innovative antimicrobials. AMR is driven by the selective pressure exerted by traditional, target-specific antimicrobials and propelled by the misuse of antimicrobials in agriculture. Both issues are addressed by the Genome Canada-funded PeptAID Project, which involves the UVic Helbing, UVic Hof, and UBC Birol labs and various partners across public health and industry. PeptAID is exploring antimicrobial peptides (AMPs) as antibiotic alternatives for application in the poultry industry. Alongside antimicrobial activity, AMPs can interact with host cells to modulate the immune response. I am investigating the immunomodulatory effects of PeptAID AMPs on avian macrophages. Macrophages are a class of innate immune cells that can be polarized along a gradient of pro- and anti-inflammatory phenotypes, depending on the stimulus, and thereby provide an informative model to assess the magnitude and direction of an immune response. My research considers the ability of AMPs to interact with macrophages and attenuate hyperinflammation caused by bacterial lipopolysaccharides, providing insight into the potential of PeptAID AMPs to fight bacterial infections. Research findings will contribute to the selection of safe and effective AMPs for in vivo trials.
